Is it ok to have about 4cm of course gravel and then 2cm of sand in the DSB... or will that serve no purpose at all?
edit: the course gravel being at the bottom.
It would not have any advantages, and could have quite a few disadvantages (depending on the size and thickness of the course aggregate). I would suggest you only use a well graded, fine sand throughout the sand bed.

Ditto. You can't keep an effective sand bed in a nano tank. Any sand you use will be aesthetic (or perhaps to create another niche for worms/pods/etc.)
